چکیده

The interactions of dehydroepiandrosterone (DHEA) and dehydroepiandrosterone sulfate (DHEAS) were investigated with various binding sites of the gamma-aminobutyric acid (GABA(A)) receptor complex to rat brain membranes, and on GABA-induced [36Cl-] influx in mammalian cortical cultured neurons. DHEAS and DHEA did not affect the binding of [3H]flunitrazepam to the benzodiazepine binding sites. In contrast, DHEAS, but not DHEA, inhibited the binding of [3H]GABA and [35S]TBPS to rat brain cerebral cortical and cerebellar membranes in a concentration-dependent manner. DHEAS decreased the Bmax values of both the high and low affinity GABA receptor binding sites without affecting their affinity constants. In contrast, DHEAS inhibited [35S]TBPS binding competitively, as analyzed by Scatchard analysis. In dissociation kinetic studies, DHEAS dissociated [35S]TBPS from rat cerebral cortical membranes in a monophasic pattern that was similar to that observed with inhibitors of GABA(A) receptors such as TBPS and picrotoxin but different from pentobarbital and GABA. Taken together, these results suggest that DHEAS binds to the TBPS/picrotoxin site of the GABA(A) receptor complex, and this interaction may be responsible for the noncompetitive inhibition of GABA responses observed with DHEAS. Furthermore, we confirmed that DHEAS inhibits GABA responses, as measured by GABA-induced [36Cl-] influx in cultured cortical neurons. Studies with DHEA indicate that this neurosteroid does not interact with the GABA(A) receptor complex.

The neurosteroids dehydroepiandrosterone sulfate and pregnenolone sulfate inhibit the UNC-49 GABA receptor through a common set of residues.

Neurosteroids are endogenous neuromodulators that bind and allosterically regulate GABA(A) receptors. Residues were recently identified in the first transmembrane domain (M1) of GABA(A) receptor subunits that are important for neurosteroid modulation. We are studying the inhibition of GABA(A) receptors by sulfated neurosteroids. One of these neurosteroid, pregnenolone sulfate (PS), depends on s...

Residues in the first transmembrane domain of the Caenorhabditis elegans GABA(A) receptor confer sensitivity to the neurosteroid pregnenolone sulfate.

The GABA(A) receptor is a target of endogenous and synthetic neurosteroids. Little is known about the residues required for neurosteroid action on GABA(A) receptors. We have investigated pregnenolone sulfate (PS) inhibition of the Caenorhabditis elegans UNC-49 GABA receptor, a close homolog of the mammalian GABA(A) receptor. The UNC-49 locus encodes two GABA receptor subunits, UNC-49B and UNC-4...

Neurosteroids and GABA-A Receptor Function

Neurosteroids represent a class of endogenous steroids that are synthesized in the brain, the adrenals, and the gonads and have potent and selective effects on the GABAA-receptor. 3α-hydroxy A-ring reduced metabolites of progesterone, deoxycorticosterone, and testosterone are positive modulators of GABA(A)-receptor in a non-genomic manner. Allopregnanolone (3α-OH-5α-pregnan-20-one), 5α-androsta...

The neurosteroid dehydroepiandrosterone (DHEA) and its metabolites alter 5-HT neuronal activity via modulation of GABAA receptors.

Dehydroepiandrosterone (DHEA) and its metabolites, DHEA-sulphate (DHEA-S) and androsterone, have neurosteroid activity. In this study, we examined whether DHEA, DHEA-S and androsterone, can influence serotonin (5-HT) neuronal firing activity via modulation of γ-aminobutryic acid (GABA(A)) receptors. The firing of presumed 5-HT neurones in a slice preparation containing rat dorsal raphe nucleus ...

POSSIBLE INVOLVEMENT OF GABAA RECEPTOR SITES IN CATALEPSY

The present work was performed to show the possible interaction of GAB A, and GABAB receptors in catalepsy. The following results were obtained: 1- Muscimol (MUS), a GABAA agonist, induced catalepsy. This effect was dose-dependent. Baclofen (BAC), a GABAB agonist, had no effect in this respect. 2- Catalepsy induced by muscimol was potentiated by pretreatment of animals with bicuculline (BI...
